چکیده
Abstract Protective immunity to cutaneous leishmaniasis is mediated by IFN-γ–secreting CD4+ Th1 cells. IFN-γ binds its receptor on Leishmania-infected macrophages, resulting in their activation, production of NO, and subsequent destruction parasites. This study investigated the role Semaphorin 3E (Sema3E) host Leishmania major infection mice. We observed a significant increase Sema3E expression at site different timepoints following L. infection. Sema3E-deficient (Sema3E knockout [KO]) mice were highly resistant infection, as evidenced significantly (p < 0.05–0.01) reduced lesion sizes lower parasite burdens times postinfection when compared with infected wild-type counterpart The enhanced resistance KO was associated 0.05) increased T CD11c+ cells from displayed costimulatory molecules IL-12p40 more efficient inducing differentiation Leishmania-specific than Furthermore, purified showed propensity differentiate into vitro, this inhibited addition recombinant vitro. These findings collectively show that negative regulator protective suggest neutralization may be potential therapeutic option for treating individuals suffering leishmaniasis.

CD25+CD4+ cells contribute to Th2 polarization during helminth infection by suppressing Th1 response development.
Mice infected with Schistosoma mansoni develop polarized Th2 responses in which Th1 responses are prevented by IL-10-mediated suppression of IL-12 production. We show that dendritic cells from infected mice are primed to make IL-12 in response to CD40 ligation, and that IL-10 acts by inhibiting this process. In infected mice, two subpopulations of CD4(+) cells, separable by their expression of ...
متن کاملSemaphorin 3E, an exception to the rule.
Class 3 semaphorins (Sema3s) regulate axon guidance, angiogenesis, tumor growth, and tumor metastasis. Neuropilins (NRPs; NRP1 and NRP2) are the cell surface receptors for the Sema3s. However, to signal, interaction of Sema3s and NRPs with plexins is obligatory. In this issue of the JCI, Casazza and colleagues report data that challenge the conventional wisdom about the role of Sema3s in tumor ...
متن کاملVaccination with Live Attenuated L. Major and TLR4 Agonist Promotes a Th1 Immune Response and Induces Protection against L. Major Infection in BALB/c Mice
Background: Toll like receptors play a major role in immune responses against Leishmania parasites. Objective: To evaluate the efficacy of vaccination with live attenuated L. major and TLR4 agonist in protection against L. major infection. Methods: Attenuated L. major was prepared by continuous sub-culturing of the parasite. A total of 90 mice were assigned to 9 groups including 6 groups of BAL...
متن کاملVEGFR2 (KDR/Flk1) Signaling Mediates Axon Growth in Response to Semaphorin 3E in the Developing Brain
Common factors are thought to control vascular and neuronal patterning. Here we report an in vivo requirement for the vascular endothelial growth factor receptor type 2 (VEGFR2) in axon tract formation in the mouse brain. We show that VEGFR2 is expressed by neurons of the subiculum and mediates axonal elongation in response to the semaphorin (Sema) family molecule, Sema3E. We further show that ...
متن کاملTh1 Immune Response Induction by Biogenic Selenium Nanoparticles in Mice with Breast Cancer: Preliminary Vaccine Model
Background: Tumor associated antigens can be viably used to enhance host immune response. Objectives: The immunomodulatory effect of biogenic selenium nanoparticles (SeNPs) was compared between treated and untreated mice with crude antigens of 4T1 cells. Materials and Methods: Female inbred BALB/c mice (60) were injected by cancinogenic 4T1 cells causing breast cancer. After 10 days, all tumor ...
